Realistically, the only way that Prince Charles will not become King is if is mother outlives him. Now, that is not such a crazy idea as Elizabeth could live on for decades, but apart from that possibility, Charles will be King. Queen Elizabeth II would never try and have her son skipped over for the throne, and indeed, I don't think that can be done. Charles has a birthright to the throne. I'm not sure what he could do (short of high treason or herresy for something like that) to warrant being passed over.
The only other realistic possibility would be if Charles were very old and incapacitated at the time of Elizabeth's death. If Charles were to have suffered from severe mental deterioration, then maybe he would be passed over.
Otherwise, get ready for King Charles III. It would probably not be a long reign, but I'm sure it will be great.
