The specific question was

How would one overexpress an important protein. Include the method used to enable rapid one-step purification.


My answer is

Isolote the gene encoding for the protein and ligate to His-tag. . Then clone modified gene into a LAC promoter in a bacterial artificial chromosome and tranform into E-coli, make sure the BAC has Ampicillin resistance gene. Then select for bacteria having Ampicillin resistance. intruduce lactose to induce LAC promoter. Then remove proteins and apply to affinity medium to which the HIS tag will bind and then wash to remove unwanted proteins. then elute wanted proteins using imidazole.

A previous answer says not to feed rasins and grapes, amongst other things. Chocolate I understand, but not rasins. My collie absolutly loves almost any fruit or veg you care to name that she has tried - although she isn't always sure what to do with the grapes.

All of us here dispute creation vs. evolution.Why is it taken seriously when Darwin himself said in "Origin of Species" that if we can't find predecessor fossils to the Cambrian fossils his theory not be taken seriously?I so far cannot find any transitional fossils that have not already been debunked.Even the former noted evolutionist George Gaylord Simpson called the sudden appearance of many types of complex life forms in the Cambrian rocks the "major mystery of the history of life.
I'm beginning to see more and more as I read along that Darwin himself didn't believe it was true.

I've held it long enough but what kind of book is the Quran? It says kill all infidels and non-believers and don't make friends with Christians (I'm one of them) and Jews because Allah doesn't guide them.

The difference between Timothy McVeigh and Osama Bin Laden is that Timothy wasn't following the Bible (at least the New Testament which we follow) and Osama Bin Laden was following the Quran.

I don't hold prejudice against Muslims and this question will probably get deleted but at least you know my thoughts right now.
