To select for transformed plants, sterilized seeds were suspended in 0.1% sterile agarose and plated on kanamycin selection plates at a density of approximately 3000 seeds per 150 × 15 mm2 plate, cold-treated for 2 days, and then grown for 7–10 days in a controlled environment at 24°C under 23 h light 50–100 μEinsteins m-2 s-1. Selection plates contained 1/2X MS medium (Sigma Chemicals #M-5519), 0.8% agar (Sigma Chemicals #A-1296), 50 μg ml-1 kanamycin monosulfate. In some experiments, Petri plates and lids were sealed with surgical tape for the first week of growth. Excess moisture during growth was removed by briefly opening the plates and shaking moisture off the lid. Transformants were identified as kanamycin-resistant seedlings that produced green leaves and well-established roots within the selective medium. Some transformants were grown to maturity by transplanting, preferably after the development of 3–5 adult leaves, into heavily moistened potting soil.

We have a library in our house. I take a book and put it on top of the bookcase thats built into the wall where no one can see it, and he comes in and tells me what book it is. How does he do this? He says it's easy.

Scientists performed an experiment to determine whether there is a connection between learning ability and food. They took two groups of 20 mice each, all from the same purebred strain. The mice were deprived of food for 3 days and then given a standard learning session in running through a maze. They were trained by giving them a mild electric shock whenever they took a wrong turn. Immediately after each learning session, one group of mice was fed, but the other was not, A week later, all of the mice were tested to see whether they could still run the maze correctly. The group that had been fed had retained this ability, but the other group had not. The probable reason that the scientists used only mice from the same pure-bred strain is so that:

a)the mice would all be the same size

b)the mice would all be the same color

c)the experiment could be repeated with the same mice

d)genetic differences would not affect the outcome of the test

e)the experiment wouldn't cost

I have a 1 inch dia. 5 foot stainless steel spit rod,a pillow block and 1750 rpm motor. need to know what sprockets to use to slow the spit down to three or three and a half rpm's.

Just a general opinion of it, whether it should be a tourist attraction in major city museums, or if you just think it's cool. Would you go to one these exhibits if it came to a city near you? If you don't know what plastination is, check out : http://en.wikipedia.org/wiki/Plastination
and then look at some of the links. I think it is a great learning tool, but some people think it is disgusting or wildly inappropriate.

How can we tell what is hot, cold or just ok?
Does anyone have an experiment of the normal temperature that people can stand? Is it 37c?
Since i was little, ive always wondered if human can stand the low temperature better than the high temperature, that i always refused my mom when she wants me to get something out of Microwave..(or if i didnt refuse, i would take a lot of things to cover my hands)

So, tell me what you think..or if you're putting left hand in microwave and right hand in the fridge, tell me whats the result.
