Due to the fact that the urinary bladder is innervated by the autonomic nervous system, and that sympathetics are responsible for increasing muscle tone of the sphincter, while decreasing the muscle tone of the bladder wall (while the opposite is true for parasympathetic stimulation) it seems counter-intuitive for a "flight-or-fight" situation to cause this. Am I missing something? Ok, obviously I am, but what?

I'm using TA cloning kit from invitrogen. The plasmid is pCR (if memory serves me properly).
I amplify the amplicon from bisulfite treated DNA, and cut my band out of LM agarose. I perform a gel purification, and perform an overnight ligation. I do a transformation the following day, and grow the bacteria out on an ampicillin plate. I see mostly blue colonies, and some white colonies.
I send out the plates (overnight delivery) to a Texas company: Seqwright. They pick white colonies (8-12) which I have circled for them and grow out in LB media containing ampicillin (maybe kanamycin, too, I not sure).
On one occasion Seqwright has been unable to grow out the picked colonies. All "inserts" were batched in the way I described above. All failed. Other batched inserts processed in the same way, are able to grow.
What did I do to that one batch I sent out for sequencing? Why did white colonies not grow out in media. (I triple checked all steps).
All advice is welcomed.

It was like a small earth worm, but it had a neon-like paint on it. It left bright neon on the grass and on my hand. When i touched it, the bright green glow got on my hand and off of the worm or grass. I took it into the light, the glow went away. but it didn't look like the glow worms you would get if you google-imaged them. Just like a worm, with neon under a black light.. PLEASE HELP ME AND TELL ME WHAT IT IS!!!!!

The term axial relates to the head, neck, trunk, and axis of the body, and the term appendicular pertains to the limbs and their attachments to the axis. Does this include body parts on both the anterior and posterior body surfaces? For instance, would the popliteal (back of the knee) be located on the appendicular body region?

Antibiotics, medication, and longer lived lives seem to be a trend during the past 50 years.

Compared to the last several thousand years of human life, no effective medication helped humans survive as such. Only the healthiest of the healthy had a chance, which meant healther offspring in the long run.

Our future offspring is "too many", in proportion to our longer survival rate. Which means we need to have less children.

We are on a path of weaker people, tinkering with Mother Nature. Sure, it normal to help the sick, but to force a brain dead siamese-twin baby to live 10 years on a machine is insane. Mother Nature would not allow that. OKAY, bad example, but you gotta see some point of what I am saying here.

At some point, our bodies may become "lazy" to heal itself. In history, the peasants were always healthier than the rich. For many reasons, such as exercise, and eating "dirty food" or "more natural food", boosting the body's immune system.
