1. Some of the circular lambda molecules are colvalently linked at the COS sites. Other circlets are only hydrogen-vonded and can dissociate to form linear molecules. Heating Lambda DNA to 65 degree C for 10 minutes linearizes any noncovalent COS circlets in the preparation by breaking the hydrogen bonds that hold the complementary COS sites together.

a. Set up duplicate restriction digests of lambda DNA with serveral enzymes. Then heat one reaction from each set at 65 C for 10 mins. while holding the duplicates on ice. Relate changes in restriction patterns of heated versus unheated DNA to a restriction map of the circular lambda genome.
b. how can the data generated by this experiment be used to quantify the appros. % of circular DNA in your preparation?

2. Design and carry out a series of experiments to study the kinetics of a restriction reaction.
a. Determine approx. % of digested DNA at various time points
b. repeat experiments with serveral enzyme and DNA dilutions.

Such as knowledge of right and wrong, a conscience, love, sense of humour, grief, emotions etc. How are these things considered to have developed through natural selection. If you don't have a sense of humour or a conscience are you a target for natural selection?

With modern medical abilities, how long will it be before they take a "sample" dna from ancient giants, inject them in a willing human participant, & grow them enough to give a pre-mature birth (giants, are bigger you know, have to take em sooner!)

Then start re-introducing giants?

Will these creatures have souls & go to heaven since they have human dna?

Do you think the white man is a modern giant? Average is 6 ft or close to it, while midgets in other countries may be 4 foot or slightly above.

Is this what the bible meant about a land of giants?

Frequency is increased for each trail. each trail last ten seconds and fatigue is shown each time (according to zero seconds and ten). As freq gets higher and higher, the tension starts declining dramatically. in physiological explanation, whys the tension declining?
