HIV and hepatitis are tested by a blood sample.
Chlamydia and gonorrhea are tested by a urine sample and sometimes a swab sample.
Trichomoniasis is diagnosed by a physical examination and laboratory test.
Herpes can be diagnosed by a physical examination, a blood, sample, or a sample of fluid from the sores.
Syphilis is usually diagnosed by a blood sample, but it can also be diagnosed by a swab sample from the sores.
HPV is usually detected by an abnormal pap smear. There is NO test to detect HPV in men.

Depends on which sexually transmitted disease you're testing for.
HIV, syphilis and herpes (HSV) can be detected via blood test. With herpes testing, you shoul use the HerpeSelect test by Focus Labs. Their test is better than old tests because it is strain specific and can differentiate between HSV types 1 and 2. The old test was weak because it gave false positives (a history of previous chicken pox infection would draw a positive on this test).
Other common STDs like gonorrhea and chlamydia are detected with urine tests or urethral swabs.
Lastly, there's HPV (human papilloma virus). There is no clinical test available for men right now, so there is no way for a man to know if he has HPV unless he displays genital warts, penile cancer or anorectal cancer.
Your primary care physician can test you for STDs. Otherwise go to a public health clinic (call your local health department for info), or try a GUM clinic. Or you can try Planned Parenthood http://www.plannedparenthood.org

For the herpes you can take the IGG test. It's the only good blood test that will check which type of herpes you have.
