I understand having a passport and various Visa applications. However, my question revolves around the any extra requirements driving would require over just flying into the various countries. What other difficulties would there be in driving such a distance across so many countries? Would there be restrictions driving a US registered vehicle through all of these countries?

Before you cross the boarder get insurance, it is the law in mexico, and make sure you have a spear tire. Hopefully you can read spanish. Go with someone who knows the way. Take change for the toll roads, and dress down. Hide you money in several places. Becareful. Especially if your light eyed. Don't look rich and dont bring a car that the feds would want to keep, they can do what ever they want to you there so make sure you have a cell phone that works in mex. I have been there a million times and there is good and bad in all countries, remember your not in the States and you have no rights there.
