The basic answer is - there is no place you can leave luggage in Manhattan near the Port Authority (if you expect to see it again) .

There are no lockers at the port authority.
If you are staying at a hotel, leave your bags there. If you are driving, leave the bags in your car. Other than that you will have to lug them around with you.
Can you maybe ship some things in advance? Other than that, make sure you have wheels and handles on your bags so that you can easily transport them and not have to carry it all on your shoulder.

My friend, there are no locker at PA Bus Terminal. If you're staying in a hotel, you can leave it at the front desk, check with the hotel before doing so. Otherwise you're out of luck.

Try local hotels or Grand Central. The hotels usually hold items for guests but may do so for a fee for non-guests.

There used to be lockers in the PA Bus terminal that you could rent. Assuming you only want to leave it for a few hours while you go site seeing it shouldn't cost much
