When you have your blood oxygen level checked, say with a pulse oximeter, it is checking the actual level of oxygen that is attached to the hemoglobin in your arterial blood. Thats why its done on your finger or say an earlobe or if a baby on their toe.

Arterial blood. Venous blood is no longer oxygenated. Arteries carry oxygenated blood to the body veins take the deoxygenated blood back to the heart to get pumped in to the lungs to be reoxygenated.

Venous blood that is carried from the body towards the heart ( basic explanation ) is lower in oxygen supply. The blood, upon, entering the lungs, is oxygenated because upon entering the lungs, this supplies the blood more oxygen to the blood flow. This blood is then pumped into the body as oxygen enriched blood ( higher concentrations of oxygen) to supply the body tissues of oxygenated blood .Again, a simple basic explanation.
