Depends what you're doing. You can actually use your computer as a preamp and send a signal from your guitar to your computer, and if you have music software you can modify your signal and send the sound out to your amp. I know of quite a few people who do this for live gigs.
Now if you're talking about recording, it's a bit tricky. If you have good mics, a good amp, and a room with good acoustics, use that route. If you don't, you can run your guitar directly to your computer and modify the sound there if you have the software. Alternatively, and this is what i do, you can run your guitar through your amp, and through the DIRECT OUTPUT or headphone output, run a line to your computer's input. Do NOT send a signal from your head that you'd send to your cab (if you have a stack setup.) My roommate fried the input on the computer i am typing this response on by doing it that way.

An amp, obviously. Unless you're got top of the line computer speakers, any $100 amp is the way to go.

Definitely an amp. Especially if you're going for the bluesy tone. Then you will need a tube amp. No way you can get that sound out of computer speakers, no matter how good they are.

It works well either way, but the tonal projection is best through the amplifier. You can play without any amplification, but that doesn't give you the sound quality that is needed for performance.
