If you're taking Amtrak straight to New York City, you will be getting off at Penn Station New York, not Grand Central. If you're going to Grand Central from Boston, the only way to do this is by taking Amtrak to New Haven, CT and then take Metro-North's New Haven Line, which is a commuter line, from there to Grand Central in NYC. There are no trains that goes to Boston direct from Grand Central Terminal. If you insist on leaving from GCT, you will have to take a Metro North train to New Haven, then transfer to an Amtrak train there.

If you want a direct train, you need to get to Penn Station on 34th Street and 7th Avenue, where Amtrak trains can take you direct from there to Boston. The flagship Amtrak Acela Express hi-speed train can do that in 3.5 hours. If you take the Northeast Regional train (the local), it can take 6-7 hours.

From GCT, take the New Haven Line to New Haven and then transfer there to Amtrak to Boston. OR....you go from GCT to Penn Station via the 42nd Street Shuttle and then switch to the 1,2,3 lines at Times Sq. From there, you can go direct to Boston via Amtrak.
