depends, if you mean handgun chambered in a handgun cartridge, then S&W does with the 500 S&W. But if you mean just plain most powerful, there are a lot of custom handguns chambered in rifle cartridges like 45-70 etc that are more powerful.

Any handgun company that specializes in single-shot pistols chambered in centerfire rifle caliber. You did not state specific criteria so I took it to the limit. .500SW is peanuts to a 30-06 Lone Eagle pistol.
