I wonder because it ends in "s," but it's multisyllable.
Would this sentence be, "Wo ist die Telefonnummer des Gaestehauses?" Would I use "von," or is that only for proper names?

In the book, it's Gaestehaus.

In general, when you have a compound word, the genetive ending is the ending of the last part of the noun, in this case "Haus", with -es.

you can say 'die Telefonnummer des Gasthauses' (not Gästehaus) or 'die Telefonnummer vom Gasthaus' - you can also use 'Gasthof' which is masculine - it might be that 'vom Gasthaus' is more colloquial, but I'd probably write it too

If you mean an accomodation for guests, not an inn, you are right with everything. If it should be an inn, it would be "Gasthaus", indeed. Yes, "von" is only used for names of persons or locations.
