Yes, your Playstation 3 has to be on for you to charge the Sixaxis Wireless Controllers. You can charge the controller 3 different ways:

You can plug the controller into the PS3 while it's on.

You can plug the controller into a laptop or computer and it will charge that way. The laptop or computer must be on for the PS3 controller to charge.

And, you can charge it by plugging it into a wall. But to plug the controllers into a wall outlet, you need to buy the wall chargers seperately.

To fully charge the controller on low battery power, it would take about 1 hour to 1 hour and 20 minutes.

Also, you can charge the controller while you play a game.

Yes it does. But the controllers charge to a good level quickly and can still be used to their full extent while charging, motion sensing and all so it doesn't really change things. A bit like the PSP.

Yes. But you can plug it into your computer's usb port and it'll charge. That's what we do so we don't have to keep the ps3 running all the time.
