For me, this is one of the most enigmatic part of the whole Bible. It is part of the whole passage where Christ answers the philosophical questions of Nicodemus, a Pharisee and a leading Jew. The more complete part of the passage goes like this: "No one has gone up to heaven except the one who came down from heaven, the Son of Man who is in heaven; and the Son of Man must be lifted up as Moses lifted up the serpent in the dessert, so that everyone who believes may have eternal life."
I think what Christ is saying is that salvation comes to the man who believes and transforms himself from the earthly to the spiritual realm and becomes a follower of Him. All these, cannot be truly understood but can be only accepted through the intercession of one who has truly come down from heaven (Christ), who is a resident of heaven, and one who is soon to ascend to heaven. Enigmatic but transfroming, Huh?

Jesus was referring to himself as the one who descended from heaven. In other words, before he became a man with human flesh and bones, he was with his Father in heaven (John 17:5) as a spirit even before the creation of the universe (Prov. 8:22-30). After his resurrection from the dead, he ascended back to heaven, the first man to be there.

Probably that no one will go to heaven until Jesus does first. There is a whole theology that when Jesus ascended to Heaven Sheol was split and part of it moved to heaven as well. I'm not too familiar with the theory, though.
