Yesterday was my 21st birthday, but it was a Thursday so I was waiting for the weekend to go out and drink and do whatever. I lost my drivers license this morning, and I have no idea where it is. Is it still possible to get into bars and clubs w/o it? The DMV is closed until Tuesday. How can I get into bars w/o all the hassle?

Youll need a form of government issued ID in lieu of the DL or ID card.
Id also recommend that you call your local police department or sheriffs office and file a report that you lost your license. Request the case number, and then contact the agency's main office the next day and obtain a copy of the report for your records.
That way if someone finds it and tries to steal your identity or whatnot, youve got some proof you reported the loss promptly

drink at a friends house instead this weekend, then go to the DMV when they are open and get a replacement. You may get lucky and find it in the mean time. Is it possible that one of your friends may have lifted it, so that they now have a fake ID?

If you have a passport, you could use that instead. If not, then there's not much you can do but get a new license.
