Always use Ms. even if you do know. Mrs vs Ms has nothing to do with your business relationship. These terms are not used in business any more.

In the professional world, always use Ms. (unless, of course, she has a doctorate, then use Dr.). It can be seen as offensive and/or presumptuous to refer to a woman in the work place as Mrs or Miss.

The preferred business use nowadays is Ms.
The only exceptions are, if the woman specificially asks that you use a different title (such as 'Miss'), OR if you know that she has a 'higher level' title, such as 'Dr'.
