Manuals are faster because they dont use power sapping torque converters that automatics require to shift the gears. As for the NOS, it is best to use it in high RPM. Right off the line, your wheels are just gonna spin and you'll be going nowhere.

Ultimately, it will depend on the specific transmission. An automatic transmission has the potential to be superior for a drag racing application as it can do everything better... but the automatic transmission you will find in any street car is not designed for that application, and as a result robs more power, takes longer to shift, and weighs more! Compared to a street variety automatic, a manual is vastly superior - though it does take more skill from the driver to achieve a good launch.
But if you were building an automatic transmission strictly for drag racing, you would not need to contend with how smooth it felt while driving on the street, if it stalled at low rpms, etc... and you could have a better system (which is what you find on most cars built strictly for such purposes).
As for NOS - the earlier the better as it provides more power... at least, as long as it's not providing more power than you can use! If you are spinning madly already, more power won't help you... in that case, as close to the torque peak as you can (but before the hp peak) when you have solid traction.

Most drag cars like the ones you see on pinks have automatic transmissions because they are primarily used in bracket racing where consistancy is key. Manual transmissions would be a faster choice. Prostock cars use manual transmissions because they are faster. The more gears you have, the better the hp and torque is applied to the ground.
Most stock production cars are geared for street use, so the gear selections may not be optimal for drag racing. You can break the tires loose at higher rpms.
Automatic transmissions use more hp to operate as well.
Some Promod cars use manual transmissions that shift with air, but are required by rules to shift them manually with the push of a button. You can set up transmissions like those to shift on rpm or time.
In general, if the transmission is set up right and the person doing the shifting knows what they are doing. The Manual will be faster.
As for the NOS, I have heard that N2O is like a hot girl with std's, you want to hit it, but you have to be careful of the results!!

I bet if you asked at the local PD, they could tell you of a drag strip near you that the police in some areas have opened on certain days or times just for this purpose. Racing down the street has never been a smart thing to do. You put the general public in great danger. Most of these kids have a lot of money to piss away, and no experience to go with it. By openong and operating these drag strips the police may stop an inexperienced idiot from killing himself or someone else. There is a lot more to consider than just your desire to go fast. Herein lies the #1 problem. You think of nothing else.

Most of the drag cars I know have an automatic transmission which is heavily modified for drag racing. An automatic is more consistent and reliable compared to a manual transmission, but you have to beef it up so it will take the abuse.

I have a super comp dragster and we use a two speed power glide transmission. I would say 95% of the cars use this transmission. Not to good for the street.
