Actually it's not the .500 S&W magnum, it's the .600 Nitro Express. It's not very common but look it up on google.

This kind of question is difficult to answer with any degree of accuracy. You see, when Dirty Harry proclaimed on the Golden Screen in 1971 that the .44 Magnum was the most powerful handgun in the world his statement was pure hyperbole. The .454 Casull, a far more powerful handgun, had been around since at least 1959. So you see, his statement wasn't all that accurate.
Today one could argue that the .500 S & W Magnum is the world's most powerful handgun, then you find out that people are shooting single shot pistols chambered for the .600 Nitro Express cartridge. So you see, this kind of question is very difficult to answer accurately.
To keep it simple let's just say that the .500 S & W Mag. is today's most powerful revolver commercially available. And the .50 AE is probably the most powerful semi-automatic pistol commerically available.

The biggest caliber handgun is the 577 Boxer.
The most powerful would be a Thompson Center Contender shooting a rifle caliber. (30-30 Winchester) or (375 JDJ).

Currently, the .500 S&W magnum is is the most powerful production handgun and round.There are other .50 cal guns, like the Dester Eagle semi auto.

Along with the DE .50 Cal there is also a .50GI.
The S&W 500 Mag is the largest production handgun, I think.
I have yet to shoot one. Something tells me it will be fun but expensive!

500 smith is a thumper and it is the largest we are allowed in the us..but there is a feller in europe that makes a single action revolver in the 600 express caliber,,but we can not import them

500 S&W but there is probably bigger blackpowder ones.
Also the 500 S&W is the most powerfull handgun cartridge avalible today.
