I think we need to know when a fight has been won before we can see what the most effective technique is. Because I have no one to argue with me over here, I'm just going to present my opinion as fact =P A fight is won when one person cannot do something they need to do to keep fighting. (Makes sense anyway, huh?) So we need to make sure our opponents can't do something they need to. That includes breathing, seeing, being conscious, moving, and attacking. So if you can keep a person from doing any of those, you've won the fight.
Each of these has a set of most-effective moves that will accomplish them.
To keep a person from:
Breathing – you need to close off their airways or wind them (with, for example, a punch to the stomach). Winding them may not be effective, however - too many counters, or maybe it just won't incapacitate the person enough. And the chest, where the lungs are, is too well protected to be effective. So that leaves the throat. As several have already said, a strike to the throat will stop a person from breathing and quickly and effectively take them out of the fight. (Also in this category would be chokes and headlocks, but I’d prefer not to cover them – most have too many defenses).
Seeing – Several people have pointed out that one cannot fight blind, and a strike to the eyes will accomplish this. However, I personally wouldn’t recommend it simply because the eyes are a small target and presumably your opponent will want to protect them. While not strictly disabling, keeping your strikes where your opponent can’t see them is effective too. (Behind their back… wait a minute, why are you attacking from there? Or low, or to the side. You get the picture.)
Being conscious – Rendering a person unconscious would be the most incapacitating thing you could do, but it’s tough to do because the body is built to protect your important bits. A hard enough strike to the head or a continual lack of oxygen will accomplish this, but it’s not really reasonable to expect to knock a fighter out with one punch (and see breathing for cutting off air).
Moving – Keep a person from moving and you can simply walk away. The most efficient way to stop someone in their tracks would be to destroy at least one of their knees. You think they’re going to hop after you on their good leg? I don’t. =) So a quick strike to the knee from the front or side would be fast and effective, with minimal potential for getting hurt in the process. Another benefit to striking the knee is that there are few things they can do to stop it. They can move their leg, and that’s just about it. And not only that, but if you keep your kick low, it’s much harder to see. Also effective is stomping on the top of the foot or the ankle.
Attacking – this is the most difficult to stop. They have four separate limbs they can attack with, and two points to attack with on each one. If you can break each arm and one knee, you’re good, but unless you’re on the ground, it’s going to be tough to stop this. If you ARE on the ground, though, and you can pin their limbs, it’s much more effective.
Personally, my preferred mode of offense would be to keep them from moving. Stomp their knee or foot and run off before they can retaliate, and you’ve quickly and effectively won the fight. (If you think it’s cowardly, you’re asking for a beating ;-) ) (sorry about the length – got going and just kept going =P)

The groin or the knee are still very good targets although most people turn instinctively away when a kick is coming to the groin so aim, speed and distance are all factors more-so for the groin than the knee. While most fighters can take a shot or they don't last long in the fight game most average type people don't take them quite so well. Not only that but I have to tell you that some of the fighters that I watch on television have horrible technique and they can't generate as much power in some of their elbow shots and here again aim is very important. You are right in that the body is pretty resilient and durable in some ways. One of the easiest ways to end a fight quickly is just poke them in the eye with your four fingers gathered around your thumb so that you strike directly into the front of the eye with the finger tips. It will temporarily stop most people or blind them temporarily and give you an opportunity to follow up with more techniques while they struggle to regain their vision.

A strike to the throat or a kick to the groin are probably the quickest ways, your right, in a real fight when the adrenalin is pumping its like you have just taken some hard core drugs so pain is reduced but a good hit to those two points will still take someone down

There is no method guranteed effective or best.Just watched a show called MOST AMAZING SHOOT OUTS all captured on tape.Cop shoots a guy with a 45 point blank in the gut guy takes gun away from cop gun jams guy then beats cop unconcious with his own gun.Cop shoots armed suspect 4 times bad guy kills cop with lucky shot to head as he falls.
Guy shoots cop point blank in the face cop wrestles bad guy to ground no martial art technique just rough house wrestling cuffs bad guy calls for back up then passes out.
These were of course desperate criminals the people I consider "real street fighters"not punk match ups most people call street fights .There is no method you can learn that will prepare you to handle real street fighters .As for the others 2 or 3 years solid hard training in any method and some guts and determination on your part will make you a winner.There is no magic bullet todays practice makes tomorrows winner

As far as the MOST effective way to take someone out in a REAL streetfight.....there is none!!! Every person reacts diferently and every situation is different. A drunk may have a higher resistance to pain than a sober person. The best thing I can say is simply don't fight the opponent's fight...pick your fights wisely.

first off, you don't punch at the throat, you either do a knife edge strike to the throat, or do a open-hand choke takedown, then from there while on the ground you can do a series of mounted punches followed by an armbar.
secondly if you're going to go for the groin, you could do a kick or punch to the groin then when the opponet bends down, knee them to the face, then follow that by a mounted punch combo then a strike to the throat.

An accurate shot to the throat or the temple will end a fight very quickly and do some serious damage which is what you are looking for in a fight situation, but I would target the throat as it is a bigger target.
