Helicopter is by far the fastest way, but also the most expensive.
Depending on what time you go, either the Hampton Jitney or the LIRR are going to be best. If you are going at off hours (ie not Friday night after work) then you won't sit in traffic on the jitney. Non-peak hours the jitney is about 2 hours, peak hours it is 2.5 hours (to Southampton)
However, if you are going at more popular times/rush hour, then you can get express trains out to the Hamptons that won't take very long. You do have to change trains, but you can get on a train that does not make that many stops before or after the switch. Most trains take about 2.5 hours (again to Southampton).

If you can fly in, that's the quickest way to get into Manhattan. The LIRR will get you there, but not in record time. If you're driving, you don't have much choice but local routes to the L.I.E. or the Southern State. I you must drive, I would suggest doing it late at night when traffic is at its lightest.

Jitney bus it's faster than the LIRR, too many stops on the train plus you have to transfer in Patchogue and Babylon.

Flying is best. If you have to drive do it very very late at night to avoid traffic.
