Most STDs can be tested by a urine or blood sample.

Some STDs can be diagnosed by simply just looking at the area (physical exam).

Others can be detected by obtaining swab samples of fluid.

Blood sample:
HIV/AIDS, hepatitis, herpes, and syphilis

Urine sample:
chlamydia and gonorrhea

Physical exam:
herpes, syphilis, trichomoniasis, scabies, crabs (pubic lice), and HPV (genital warts)

Swab sample:
herpes, syphilis, gonorrhea, chlamydia, trichomoniasis

Pap smear: HPV

Those obvious answers ARE the ways. A doctor may make observe what's going on and make an educated guess, but her or she will still draw blood and test for STDs. The gold standard for diagnosing herpes, or HSV, is to take a swab of the fluid that is inside the blister and culture that...but it's usually diagnosed through a blood test, as conditions are not always conducive to a good culture.

Scrape off the scab (or collect some yellow liquid from the sores) and get a doctor to analyze it.

Urine / blood tests are the ONLY definitive ways. Put your pride (and dignity) on the shelf and go and see a doctor before your situation gets worse or you start infecting other people.

Well, if you want to be a smartass, you can say "Only a doctor can diagnose an STD".

How about that?
