In the blood bank, before we release a blood for transfusion for a specific patient , we do blood typing and also blood cross matching.
What does blood typing do? to make sure that both the donor and the recipient are of the same Type- 'A, B, Ab, or O. Whether they are Rh + or -.
What does cross matching do? Each bag of stored blood will have to be cross matched to the blood of the patient before transfusing it. (This is to double check for minor incompatibilites between donor and recipeient).
The blood typing and crossmatching should be declared compatible before one bag of blood is considered transfusable to a patient.
There are diff. stages in the cross matching test:
Donors RBC vs. recipient plasma
recipient RBC vs. donors plasma
The red blood cells (RBC) should be purified. meaning there should be no plasma attached to it, or this may affect the tests. Therefore the RBC should be washed repeatedly with normal saline solution to purify it. while it is waitin for crossmatching, it is in the form of -------
RED BLOOD CELL SUSPENSION, especially when it is left to stand in the test tube rack.

a correct red cell suspension would allow you to have the proper ratio between blood and reagent when you're doing blood bank testing. it is very critical to have the right percentage of suspension because a diluted or concentrated suspension could give you false positive or negative results or in most cases, agglutination reactions. different reagents would require different percentages but mostly 3%. gel methods for antibody screening and detection generally require 0.8%

Red cells are used to detect blood group. RBC have a quoting of an antigen. Different sera with different antibodies react with RBC and depending on the agglutination ,Group of RBC is recognised. For example anti A serum found in B group blood agglutinates A group RBC..
