Several ways exist to become a professional guitarist. Play in a band, learn from other guitarists, and focus on your weaknesses and improve them.

With many players, it takes years of playing to be able to play a tune after hearing it once. Most others use sheet music to play a song.

You become pro as soon as you are being paid for your ability on guitar; whether that be teaching, playing gigs, being a session guitarist, or making albums people buy etc.

A guitarist who knows only a few chords could also be a pro. But I would guess you are talking about a reputable famous guitarist type person? That is hard to answer what needs to be learnt, cos all pro's have different talents. The most obvious things I can think of are: knowing where all the notes are on the guitar fretboard, the ability to improvise over at least basic chord progressions, have good relative pitch ear training, knowing at least all the basic chords, having the ability to sight read, possibly knowing the basic scales but this isn't necessary since some famous guitarists don't know them.

What skills you need to learn depends from guitarist to guitarist depending on what kind of music they want to play.

Lots of practice and tremendous amount of talent. As for tuning in different ways you can get a book that tells you how to do that at Sam Ash. As for listening to a song and being able to know the notation or tabs for it that's called perfect pitch those type of people can hear something just once and be able to play it back to you. That is a gift from God, I wish that I were that talented.Jacqueline du Pre the great cellist was able to do that as a child, and so could John Williams the great guitarist, Jackie at thirteen could listen to a concerto once and play it back, not only do some of them have perfect pitch they also have a fabulous memory for music, how I envy them. It takes me months to work on and memorize a concerto. Mozart was also able to do this as a child.

I learned how to tune it by ear. I'm 12, and it's not far off.
If you do TAB, you're not pro. TAB is for immature idiots who would rather impress their friends quickly than actually learn the notes or chords.

P-R-A-C-T-I-C-E is a little word I learned once.
