You can catch a Greyhound bus at Port Authority, which is on 42nd Street and 8th Avenue. (Penn Station is on 34th Street, so it's pretty much within walking distance.)

Greyhound is located at the Port Authrority Bus Terminal. If your carrying lots of luggage, and don't mind wasting $2.00 for a one stop subway, its only one subway stop uptown. Any line, 1,2,3,A,E,C all stop in Penn Station, and they all stop at the Port Authority. My recommendation is the A,C, or E. Getting off any of those lines at the Port Authority stop takes you right into the buidling. The 1,2, or 3 require some walking underground, and its a lot of ramps and stairs, and if its a hot, humid day, being in those tunnels can be quite unpleasant.
If you don't have a lot of luggage, or just feel like walking, walk uptown 9 blocks along 8th Avenue. You can't miss it, the Port Authority is a large building. Wikipedia states that it is "between 8th and 9th Avenues and 40th and 42nd Street at 625 8th Avenue."
Its an interesting building with buses that actually drive above restaurants if you look closely enough. Theres intricate pipe (only way I can describe it) work on top of the building, that defines it as the Port Authority. You cant miss it.

Port Authority on 8th and 42nd. Penn is on 8th and 34th. So, it's 8 blocks, which is less than a half mile, and would take 8 min. to walk. go to greyhoun.com
