lots of cardio and weight training. one day a week for resting.
find calorie/fitness calculator on the net, enter all your stats and it will tell you how many calories you need a day to maintain, take off 300-400cal off that number and thats how much you need to lose.
dont try to lose more than 1-2lbs a week - its not healthy to lose more.
eat 5-6 small meals a day (eat every 2-3 hours)
5-8 servings of fruit and veggies a day
8 glasses of water
have complex carbs for breakfast - they give you energy
have lean meat (protein) for dinner - repairs muscle
cardio exercise 4-6 times a week for 30-50min, light weight training
dont consume foods that are made of white flour (white bread, cakes, past etc.), sugar loaded foods (cookies, icecream, candy etc) and nothing fried - no junk food.

If you're fairly active throughout the day, carbs are a very important part of your diet, and they should NOT be eliminated or even cut back on. Obviously, you don't want to eat pure sugar, but pasta, fruits, whole grain cereals and bread are healthy carbs that will keep you energized.
Now, to cut fat without losing muscle mass: Cardio. Just do 20 minutes, 3 times a week (with a 5 minute warm up and a 5 minute cool down). Truly, the only way to legitly get rid of it is to burn it off. And the cardio can be anything... Dancing, playing tag with your kids (if you have them), running, bicycling... Do what's fun for you, whatever will get your heart rate up. Good luck!

Actually, if your carbohydrates are too low, you will lose a great deal of muscle regardless of your protein intake and here is why:
http://www.spartafit.com/articles/requirement%20for%20calories.html
The best way to reach your goal is to eat 6 small but balanced meals per day. Your carbs should be high in fiber, your proteins should be lean, and your fats should be sparing but high in the omega 3 & 6 fatty acids. Avoid greasy foods, alcohol, and candy.
Lift weights or perform power yoga. These are resistance exercises which will increase your lean mass and burn extra calories 24 hours per day. Also, some cardio helps.
Use a measuring tape instead of a scale to measure your progress because the measuring tape is more accurate.
Here is a sample meal plan and a sample workout:

Both of the first two answers are good but the bottom line is to increase your RMR, resting metabolism rate, that way you keep burning fat all day and night, yep even while you sleep. Regarding colon cleansing try fasting it is a good detox but take it easy if you haven't tried it B4 especially if you are diabetic, would pay to ask your GP (doctor) There is plenty of good info on Google
