Yes, I would put the tile under the vanity and then mount the vanity against the wall and screw it into the wall studs to secure it.
As for the tile floor, I would remove the old floor and install a new concrete backer board for the sub floor and tile over that. This will help the floor from moving and cracking the grout line. Good luck
