How old is he? What is his living situation? It could be a relative or a friend of a relative. It could be a roommate's girlfriend. Or it could be his friend, who happens to be female. There are too many possibilities. I suggest that if you do anything, you ask your boyfriend who it was.

Well you can't jump to conclusions because he's staying with his mom now so that could've been anyone. You also cant let it run your thoughts either. Don't stress over something so little. If you call and she answers and is rude once again, ask him non chalantly, "So I called you your moms for you and some lady answered the phone and she was pretty rude, who was that?" Give him a chance to answer, it might be just a relative.

DON'T JUMP TO CONCLUSIONS. First, find out who she was (in a casual voice tone). Then, give your boyfriend plenty of opportunity to explain without making him feel like he's being interrogated. HEAR what he says, don't hear what you are afraid to hear. Then make a conclusion based on what you get out of the situation. If you jump the gun and accuse him when it was simply a female friend or relative, you will lose his trust.

Confront him. Break up if he has no good explanation. She would have to be pretty comfortable to answer the phone. So she is either a close friend or relative ( in which case you should know about her) or he is playing you.

If I was you I would just simply ask him about it. I doubt he is cheating because he would have never let the girl answer. It may have been his mother or one of her friends. I wouldn't worry about it and just confront him on it maturely.
