To what end are you asking your question? The .500 S&W is more potent than the .50 Action Express, but the .45 acp & .9mm don't even belong in this category as neither of the latter are hunting rounds. What do you want your handgun for? The .45 & .9 are good personal defense weapons against 2-legged predators. The big fifties are good against 4-legged predators. The .500 S&W is a revolver round. The .50 Action Express is a semi-auto round with a rebated rim. What do you like better, revolvers or semi-autos? Overall, the .500 S&W is the most powerful of the ones you've mentioned, but what is your application?

The question is what do you want the round for... The .500 S&W outperforms the AE by far; however, for large cal hunting pistols you also have to look at the .460. It is an amazing round. I personally can not stand the 9mm round because of personal experience overseas. The .45 is my favorite overall round for all purposes, except for penetration. For penetration I prefer the .40 or the .357. The .40 is a great compromise. Still for home protection and carry I prefer the .45.

depends on what you want the firearm for. the .500 is a good hunting round. The .45 is more powerful than the 9 mm

All around "best" choice.......45. It's reasonably priced to shoot, accurate, has terrific stopping power against soft targets and there are all kinds of guns & ammo out there to suit your particular need. Sure, it hasn't got a great deal of penetration when it comes to hard objects, but how often will you shoot someone through a car door? And if they are wearing soft armor, it's gonna hurt like hell.

.50 caliber rounds are for hunting, they are impractical self defense cartridges. .45 has more stopping power than 9mm, but neither will penetrate vehicle bodies.
