It would be a fantastic trip! Not cheap though.
There are two ways, Via Rail, which goes from Vancouver to Banff, then you could get off in Banff and take Greyhound to Calgary. http://www.viarail.ca/

Or, you could take the Rocky Mountaineer from Vancouver to Calgary, might be more expensive, but it's a dedicated vacation style train. http://www.rockymountaineer.com/our_trains/rocky_mountaineer/
I've not had the pleasure to take this rail trip, but have heard from close friends and relatives how lovely and astounding it is. I have had occassion to drive through the rockies numerous times, and that alone tells me it would be well worth it!

I live in Calgary and this is some of the most amazing scenery you'll ever see, going by rail from Vancouver through the Rockies. I'd advise going as soon as it warms up, most ideal time being the height of the summer, June through September! Fall is awesome too! April and May are not bad either, but the fullest splendour would be in the heart of summer! Unless you want to see snow, then by all means, any time during the winter months as well. It's always breathtaking, any time of year! Have fun!

You can contact Via Rail.com
There is no longer a passenger train route into Calgary
but you can go from Vancouver to Edmonton
The weather does not matter on that route as is travels
through the Rockies and is awesome ANY time of the year
Trips cost from $ 487.00
Very expensive way to travel but would be worth the money

A worthwhile trip in general. Try to do it in summer, when there is a greater amount of daylight (until 10 pm in early July). Also, that time of year is when almost every place has a variety of annual festivals and events going. Especially in Edmonton if you go that direction.
