My favorite restaurant in Regina is the Lakeshore. It's sort of a steak place, but they have lots of other things on the menu. The service is phenomenal (they have old-school french waiters in suits, very formal), the food is fantasic. The prices are a little high, but only by Regina standards.

I honestly couldn't commend on wheelchair accessibility since I have no need for it myself, but I'm reasonably certain there is a button for the door, and there are no stairs or anything, so it should be fine.

My second choice is Poverinos. They make some really interesting pastas. Service is hit and miss - it's mostly college students working. The prices are pretty decent, ~$12 entrees on average sort of thing. Again, it's a single floor place and there is a button for the door, so a wheelchair shouldn't be a problem.
