The onset of action for both Lisinopril and HCTZ is about 2 hours, and their peak is at about 4-6 hours. That means if you actually sit and check your blood pressure every 10 minutes after taking the pill, you should see it starting to decrease after about two hours. However, it will probably be a couple of weeks to a month before you have a consistent decrease in your blood pressure throughout the day because it takes several doses for the concentration of the medication in your bloodstream to reach a "steady state".

In theory, it is probably best to take the drug first thing in the morning because our blood pressures tend to run highest around 8am and 6pm. However, some people may feel some lightheadedness when standing from a chair a few hours after they take their medications when their effect is at its peak. In these cases, we usually tell patients to take their meds at night so that they are lying flat (and presumably sleeping) during the peak effect.
