The most used language in the businessworld/people/etc.. is Chinese (specifically "mandarin").
However, in most of the anime/manga series: Japanese.

Chinese is the most spoken language in the whole world.
And for you other question...I believe most anime/manga is in Japanese as is originates from Japan, I think...and the ones I know of all speak Japanese...

There are many different languages in China. The two major ones are Mandarin in the north and Cantonese in the south but there are a dozen or so others. China has ten times the population of Japan.
