My husbands sister only calls him when she needs something. Take my kids here, grocery shop for me. It gets very tiring after awhile. And then his brother is the constant 1 upper. He does everything better and he's so much better at everything and he knows everything. I've tried to tell my husband to stand up for himself but he's like a doormat. It's starting to wear on on marriage. Do I give up and let him be a slave or try to get him to realize he doesn't need to do everything his family wants.

Don't come between family you will be left out in the cold. but if he complains about doing things for his brother and sister then tell him he has 2 choices: 1) Stop doing it 2) If he won't stop doing for them, stop complaining to you because you don't want to hear it

Tell him you're going out but he can't go, he needs to stay home in case his sister calls or his brother calls. Tell him you know he just lives to be run over by his sister while his brother who does nothing for her brags about how smart he is. And you wouldn't dare think to have him leave the house in case he might miss something with these 2.

why don't you let him do all that and some. If he likes to do things so much for his family, that means he needs more responsibility. try giving him more chores around the house and be more with the kids that should tire him out.
