No... do not defrost in hot water. The concern is that the hot water will encourage bacterial growth. The danger zone is any temperature between 40F and 140F.
If you have some of the meat defrosted, just take that part off and leave the frozen core (wrapped in a ziploc) and place in cold water with the faucet running at a thin stream.... Also, try to break the frozen core into smaller bits so it will defrost faster.
The running water will help defrost the remaining meat quicker than just placing in cold water alone.
Also, you can use the defrost in the microwave... but use sparingly.

You did not say how much hamburger is still frozen. You say you have to get this in the oven by 4pm. I don't know what time zone you are in so here goes...
If all of the hamburger is still frozen like a rock you might want to employ two plans of action. Use microwave on defrost and scrape away thawed portion and nuke the frozen portion. If on the other hand you have a chunk that is still frozen in the middle you could put in a freezer bag (Ziploc) and hold it under warm running water. It is nearly two pm on West Coast and the microwave method would thaw things out in time for you to put meat loaf in the oven.
Also, you could mix and stick small frozen bit in corner of the pan when you put in the oven. It won't have as much seasoning but as a good host of family member you could serve that section to yourself.
With all that said I would try both microwave and hot or very warm water running over hamburger in zip-loc bag.

NOT in hot water...you use cold water or you can actually start cooking the meat.
Make sure it's in a water proof container or baggie when you soak the meat so the water never actually comes in contact with the meat.
You could also defrost only the frozen part in the microwave. Pull off the thawed portion of the meat and put it into your meatloaf mixing bowl and put the frozen part in the microwave on defrost.

When you defrost meat in water, you use cold water. I wouldn't use that method for hamburger. Remove all the thawed hamburger from around the frozen part and zap the frozen part in the microwave for 10 to 15 seconds at a time until you can work with it. It shouldn't take long. Each time you check it, remove the thawed portions. This should be faster than water.

defrost in cold water as warm water can breed bacteria in the uncooked burger. If you have a microwave that will be faster still. Also try breaking up your burger into smaller pieces as they will defrost faster in smaller chunks and letting it sit for a few minutes and breaking it up more I have used that a few times to speed up things. I wouldn't. It will not hold together! Just pull away the defrosted hamburger and the other will not take too long!
