Do they drive?
If they don't then London will be more convenient. Plenty to look at and do, and plenty of public transport to get around. Me- I can't stand the place, but its OK for a break.
If they do drive, then Bath is absolutely perfect. To be stuck in the city for more than a day or two could get a bit dull, but there is so much gorgeous countryside around and about that it is worth going down there for that as well.
You could also consider Oxford and York as possibilities if they like cities.
If they like the countryside and walking - then either the Lake District (as mentioned by someone else) or the New Forest.
or if they like to be by the sea - go to some of the little resorts along the Jurassic shoreline of Dorset - Lyme Regis, Durdle Door, Lulworth Cove are all lovely.

do a bit of verbal fishing and see if you can find out a place special to them both. London is a great place for a city break - but they may have fond memories of somewhere a little more quiet. My parents had their first holiday together in Canterbury - so my sister and I arranged a luxury weekend there for their 25th annv.
They loved it!

Windermere in the Lake district, they can walk, have tea and cakes, row on the lakes, theres even a few places of interst where famous people have lived......look up the area on the internet. great place for all ages.

Lake District or N.Antrim coast which includes Giants Causeway.If something more urban try Edinburgh.
