Depends on your tastes, budget and your expectations.
Killarney is a better place to base yourself if you don't have transport; it has all the tourist and traveler resources, the lakes, mountains castles and a lively city center an night life.
Tralee is a great place to base yourself if you have a car and prefer to visit the fantastic coastline and Dingle Peninsula (unmissable in my opinion). It is less populated, less busy but arguably more authentic in certain ways.
To answer your question, my advice is Killarney. But if you more than a few days consider both.
If you care to post more about your preferences I'll try to answer in greater detail.

Take my advice and go to Killarney. It's a much nicer town than Tralee and it's close to Kenmare, a gem of a town during the summer months when it's bussing with tourists. You are only 30 minutes or less from Tralee if you want to visit. While there don't miss the chance to brouse through Quills wollen mills and tour the grounds of Mucross House before travelling to Kenmare via Molls Gap. The scenery there is stunning. Also Killarney is quite close to Cork city if you want to do some serious shopping while there.

Killarney is more popular with tourists, there's definitely more accommodation (pricier than Tralee though), it's nearer to West Kerry, where the scenery is amazing. The nightlife in Killarney is also great. Tralee can be good for shopping and nightlife, but there isn't really a lot in the way of tourist attractions. However, whichever you decide, there's a fairly good bus service between the two towns, so you needn't miss out.

definitely Killarney. It has more charm and is prettier. It has a national park nearby with the famous lakes and walking trails and Muckross House visitor centre. Tralee has a big indoor pool and slides etc if the weather is very bad. It is not far from Killarney. Killarney also has some really nice Irish country pubs.

KIllarney is the tourist destination, Tralee is more of a working town. If you can travel, spend a few days in Dingle, nicer than Killarney IMHO

Killarney is full of tourist haunts.
if that's your thing, go for it.
I prefer Tralee.

Go for Killarney, it's more central, there is more, to do and it is geared for tourists.
Absolutely beautiful scenery, breath taking, I mean it.

They're only a few miles apart. Depending on the time of year you go there, if you don't like one, you can check out of your accommodation, get on a bus and go to the other and find accommodation there in a matter of hours!
