1. Their real-time shields should be ALWAYS be on.
2. Set them to auto-update the virus & spyware definitions. Usually this will be done silently in the background when defs are available. But if your programs require you to schedule a time for it, do it daily at some specified time when it won't interrupt your work.
3. Do scheduled full system scans for both antivirus and antispyware nightly while you sleep. (Don't set both to go off at the same time -- leave enough of a gap between them so one can finish before the other starts.)

if you really want to keep the BEST virus protection, then EVERYDAY. But most average people update it weekly
