Get some window clings and let him decorate the car windows. You can get them at hobby stores or even grocery stores. Mostly they are just holiday themed so now you'll probably just be able to find Christmas themed ones, but sometimes you can find cartoon characters. They're like $2.00 a sheet and you get maybe 10 or 12.
Bring along plenty of books for him to look at as well. Maybe books that have the buttons on the side and make noise.
Plenty of little snacks as well. Little zippy bags of goldfish crackers, pretzels, small cookies. Things that will keep him occupied but won't fill his belly up.

**Markers( now they have ones that you can draw on windows with if you are okay with that, or the color wonder markers that don't leave any marks except on the special paper).
**Get assorted little toys, books, treats, etc. that you know he'll enjoy and wrap them each individually and then throughout the trip surprise him with a new toy. It's incredible how excited they get opening these little trinkets! I'd recomend going to Target, Walmart or the dollar store.
**Bring snacks along to keep your kid happy, maybe even some treats that he doesn't usually get to have. We like to keep some dum dum lollipos along since they are so small and our girls really enjoy them as a special car ride treat.
**For the video player try bringing along new videos for him to watch that he hasn't seen before.
**If you have an old discman or tape player that you can hook headphones into that he could easily work and if it broke it would be okay that would be a good idea also.
We stopped a lot when we drove to the East Coast from Chicago to visit family, even if it was just to run around at a park for a few minutes. It kept our girls happy and we all got to stretch our legs for a bit.
Have a safe trip!

Pack a pillow, and drive at night.
I don't have a toddler, but I have a 7 month old baby with plenty of energy.
We recently got up and on the road at 3:00am. Baby-girl slept for all but the last 45 minutes. We got to my parents house at 8:30, just in time for breakfast.

We just took a long trip with our 2 year old. I bought brown lunch bags and wrapped "presents" in bags that I sealed with fun stickers. Every so often, he got a "present" to open. Didn't really matter what was inside: a single size box of cereal, a small new book, a lollipop, some stickers, a box of animal crackers, a little car. He loved it!

We travel alot and we have two kids. Since they were young I have always had a back pack attached to the back of our seats, filled with car games, toys from our dollar store ( so if you lose them it is easy to replace them again), books, and felt story board to play with. It keeps them happy. We also invested in a car TV , that sure helps too.

Obviously nothing smaller than an elbow...(from getting in the mouth) magic doodle is fun..they draw and then magically erase and start all over, figurines, dinky cars, dolls. The best thing though after all this stuff is parent particapation. I spy games.... reading books or in this case looking at picture books, singing songs...hope i was usefull :)

Books or one of those travel size doodle pads. (kinda like an updated etch n sketch). A portable dvd player to play movies has helped us out greatly on planes as well as car trips.

I'd bring lots of toys, snacks, and things that your child enjoys. I just had my first trip with a baby last weekend. I went to Indiana- a 4 hour drive from where I live, and I just took toys to play with, snacks for him to eat and lots of juices. He was very good and kept ME entertained the entire time.

Bring a graded reader, like one from the Illustrated Classics series and read to him. You can sit in the backseat while you read so he can see the pictures. Also, think about some books on tape.
