I need it to detect the signal via the parallel port on my pc...

It depends on whether you simply want a boost in voltage value or a boost in signal power. In order to simply boost the voltage you need a transformer but if you want the signal the be boosted in power also you what you need is a simple amplifier circuit.Amplifier chips are available in IC patches

It depends on what is supplying th 220mV and what you are using to detect it. The problem with a transformer to boost the voltage is that it will transfer whatever lthe impedance of the measuring or detecting device is to your 220mV source, and that could adversely affect that circuit. In your case, your transformer turns ratio would be 5/0.220 = 22.7. The impedance ratio would be this squared. or 515, If your detector has an impedance of R, the load on your circuit woul be R/515. A better way to do this would be to make a small transistor amplifier with a gain of 22.7. You can design that so that the load on your source is minimal.

Transformer. Voltage 1 Amp 1=Voltage 2 Amp 2. Find a suitable stepup transformer.

One of the options is to use a step-up transformer. Another option would be to design a transistor amplifier that has a gain of (5000/220). You can do either of this. But your question itself is insufficient. It depends a lot on what purpose you are going to use it for!
