Yes the incline will help as gravity helps to increase the weight load working against you.
The absolute best method for burning fat the fastest is cardio excercises. "targetted" excercies can only be used to build muscle, but contrary to what many apparently believe, you cannot target fat areas. Fat is something you lose overall, and your best bet for burning it is general cardio excercies like the treadmill, jumping jacks, excercise bike, etc.
In addition, it builds on your endurance faster, meaning that you are reaching a point where you can step it up more productively.

hi ya i have a similar problem i hate to run but i need to burn fat so here's what i do i put the incline up to nine or ten and power walk on a speed of 5 and that is working for me so far iv lost 10 pounds in 5 weeks so give it a go hope this helps

An incline is good. It uses more energy. Changing from flat to incline and back is good too. When you do an activity that is at an even pace, your body tries to find a conservative burn point, or plateau of fat burning. If you change your pace now and again, it keeps the work out more productive.

If you keep your running speed constant then increasing the incline will burn off more calories. You don't want to lose weight too quickly though because this can cause you to reduce muscle as well as fat. It might also reduce your metabolism and increase your appetite which will make it harder to lose the weight.

You bet it will.
You wantr to lose fat the fastest way is no carb. No bread, rice, pasta, or potatoes. You will drop it like it's hot.
Problem with any diet is if you fal off your diet you will gain all that weight back plus more.
Running uphill is harder so it burns more energy / fat

If you run down an incline you are pushing your muscels harder than running on a straight surface or running downhill. So it probably does help but don't just run up inclines do regular running too.
