I have travelled from Kanyakumari to Rameshwaram some time back. The trains are:
Rameshwaram-Madurai &
Madurai-Kanyakumari
This is the only route-recently Rameshwaram to Madurai has been converted to broad guage,
perhaps now there is a possibility of direct bogies linking these two important stations in Tamil Nadu

Right now, the rail lines from Rameswaram-Mandapam further down are under repairs/re-laying: so, you do bus or car (ie., by road), from Rameswaram to Madurai or Manamadurai, and, catch the train to Kanyakumari from there(via Thirunelveli). When the rail lines are restored you do rail travel from Rameswaram to Kanyakumari, via Madurai.
