Depends on what you're testing for. The urethral swab the other fellow mentioned is not necessary for most part. Gonorrhea and chlamydia can be detected with a urine culture. They try to see if any bacteria are living in urine (therefore also living in and around your urethra). Results take 3 to 7 days.

If they are testing for HIV, herpes and syphilis, they are testing for antibody reactivity. If antibodies react, then you currently have or previously had an infection.

The fastest HIV tests can give results in 20 minutes. This is the OraQuick rapid test.

Herpes is tough to test for. Focus Labs offers the HerpeSelect test. It's strain specific, so you won't get false-positives if you have had chicken pox. The old test wasn't as accurate. Some labs will only test liquid from fresh lesions, but HerpeSelect tests blood even if you aren't having an outbreak. Testing for herpes is really expensive, so most doctors will just go with a clinical diagnosis (see a lesion, you've got herpes!).

Syphilis is a simple blood test. Results in two weeks.

Apparently for guys they put a long q-tip in that part of your body, apparently very painful, and you get the results back in about a week.
