they will insert a small Qtip like swab inside the opening of your penis and then send that to the lab to check for any signs of infection and certain types of sexually transmitted disease's. Blood test usually checks for syphilis... Gonorrhea, herpes, chlamydia are usually checked by swab testing. HIV-Aids is checked by blood tests. So I guess you would have to know what STD they think they have,,,

It depends on what you have or what they are testing for..... If you are having discharge from your penis other than urine, then yes they will have to swab the inside of you penis...
