Just ask your doctor to test for STD's they will know what you are talking about. There are numerous tests, some blood tests some smears (where they take samples and try to grow the bacteria). If you want to be completely checked out just tell your doctor you want to be tested for all STD's

It depends on what STD they are testing for. For syphilis, they usually do a VDRL. For chlamydia a culture of tissue, There are a couple for HIV, gonorrhea is also done from a culture of the secretions,
