Squats are the best exercise you can do for an overall workout. They do work your bottom, legs, and lower back, but also get the hormones going that help in the rest of your body. Every workout plan should include squats.
Other exercises that will help are lunges and leg press. Also, walking on a treadmill at higher incline will help. When you do this, tighten your glutes (butt) while you walk. You'll really get a good workout this way.

Yes they do. Squats make the gluteus more toned and that lifts the butt. Any exercise that works that muscle will increase the size of muscle therefore the butt. Lying down, doing leg lifts may help the gluts and the abdomen. Standing and lifting the leg up and back may help as well. So go on-line and search for gluteus exercises, you'll find more.
I understand, a flat a....is less attractive.

You cant change your genetics, but building muscle in your butt can make it rounder and more toned. One thing that can give your butt a lift is this exercise: Stand behind a kitchen chair, holding onto the back of the chair. Slowly lift one leg up behind you, keeping your knee straight, then lower your leg slowly. Do 10 reps then switch legs and repeat. Squats will also work, but they make your thigh muscles bigger too.

Squats are probably the best exercise to build your glutes (butt). Some other good exercises are lunges, deadlifts, good mornings, and hyperextensions. These exercises will tone your glutes and if your butt gets any bigger as a result of these exercises, it will be from the result of muscle hypertrophy (growth).
Your butt will get smaller as you lose weight because you will lose the bodyfat from that area of your body.

Sitting for 8 hours in a job such as a secretary, can certainly make it bigger, however, I would recommend using a weight and doing the forward and backward leg stretch as one exercise. Squats will make your backside tighter, but not necessarily bigger.

Squats and lunges are two of the best exercises you can do for you bottom. They don't make it "bigger" - it's more of a "lifted" look. Just be sure you have proper form when doing them so you don't hurt your knees.
