You can hit the wineries along Rancho California Rd. in a day. They're mostly open from 10am to 5pm. I have set a record with 8 wineries in one jaunt.

Thornton is a great one (they specialise in sparkling wines). Also nice are Stuart Cellars (fabulous reds) and South Coast Winery.

One place to visit is Historic Old Town Temecula
It's an Authentic old western town over 100 years old with 350 arts/crafts and antique dealers and restaurants.
There's a tour of the wineries around Temecula, but the only one I found is a shuttle bus trip that goes to 4 different wineries and takes a full day.

Thorton Winery has a great restaurant, it a bit pricey but very good.
