He gets frustrated with himself and feels if he talks to women he will say something stupid and sometimes gets "depressed" and thinks he will end up like the guy in "40 Year Old Virgin" - Any suggestions on how to help him or advice to help him not get frustrated?

Make it a job to practice. If he wanted to learn astronomy, or martial arts, or auto repair, he has to put in the effort to try to learn.
