It really depends on what you like!
Venice, with all its canals and the Bridge of Sighs, is supposed to be incredibly romantic.
Rome is steeped in history (think Coliseum=Gladiator and very well-preserved Roman ruins) as well as fantastic art. Michelangelo is a HUGE draw for me and Rome is where you'll see his Sistine Chapel ceiling and the Pieta (in St. Peter's Cathedral).
Florence was Mike's hometown. The Galleria dell'Accademia (next to the School of Fine Arts) holds the statue David, two Slaves and another Pieta in which the artist sculpted himself. At the nearby Uffizi museum is one of the best collections of Renaissance art in the world (including Botticelli's Birth of Venus and Primavera).
I haven't been to Venice but luckily have gotten to both Rome and Florence for four-day weekends.
I think I'd return to Florence over Rome simply because the city center is more compact and, possibly because of the student influence, the city seemed more lively than Rome.
Oh yes, and if you want to kick back and see some incredible scenery, take the SLOW bus (about one and a half hours) from Florence to Siena and back. The countryside is breathtakingly beautiful and over each hill, you'll see THE villa you just have to have!

I think that Venice is great for a weekend in Italy. It's easy to get to, relatively small for a city and very unusual.

rome rome rome......went last year and had some of the best food and most relaxing time there ever. Go into an empty restaurant and let them serve you whatever they want, and let them bring you their wine selections....
all of the sightseeing there is close to each other and is beautiful all year round.

I would go to Venice, definitely...a lot to see and do and you can pretty much do it all in a weekend! Plus the food is AWESOME.
