The antibiotic starts working almost immediately. But what I think you're asking is how quickly do you see an affect. If I'm right, then . . . you are still infectious after getting the antibiotic shot. I'd wait until all lesions disappear, and probably a week or two more just to make sure, before you have intercourse, or else you could pass it on to someone else. Also, if you have syphilis, you should get tested for all other STD's, since people are frequently infected with more than one.
